of Thesis Presented to the Graduate School of the University of Florida in Partial Fulfillment of the Requirements for the Degree of Master of Science THE ROLES OF ZINC AND METALLOTHIONEIN IN HEPATOCYTE ANTIOXIDANT DEFENSE By Louis Lichten August 2004 Chair: Robert Cousins Major Department: Food Science and Human Nutrition Zinc and metallothionein are involved in cellular antioxidant defense, but the basic mechanisms are not known. Zinc is a biologically redox inert metal, that has been associated with protection from various oxidant producing species. However, zinc induces the cysteine-rich, metal-binding protein, metallothionein, through interaction with the transcription factor MTF-1. Metallothionein may have a role in preventing oxidative stress, by quenching free radicals and other oxidants. Therefore, it is hard to distinguish between protection due to zinc, and protection due to induction of metallothionein. Hence, metallothionein null (KO) mice and metallothionein transgenic (Tg) mice provide a unique opportunity to study altered states of metallothionein expression. These mouse models were used to determine the effects of zinc supplementation and/ or metallothionein gene expression on the susceptibility to oxidative and nitrosative stress. Metallothionein KO mice were significantly less viable than control (CK) mice after

Differential Gene-Expression of Metallothionein 1M and 1G in Response to Zinc in Sertoli TM4 Cells

Background: Zinc (Zn) as an important trace element is essential for testicular development and spermatogenesis. Molecular mechanism of Zn action in the reproductive system may be related to metal binding low-molecular weight proteins, metallothioneins (MT). Our objective was to determine the effect of Zn on two important isoforms of MT, MT1M and MT1G genes expression on testicular sertoli cell...

Effects of in ovo Injection of Zinc Acetate on some Gene Expression Associated with Embryonic Growth and Development, and with Growth and Carcass Characteristics of the Resultant Chicks

This study was conducted in two steps to determine the effects of in ovo injection of zinc acetate (ZAC) on some gene expression associated with embryonic growth and development, and with growth and carcass characteristics of the resultant chicks. In the first step the effect of in ovo injectionofZAC on the expression of insulin-like growth factors (IGFs:IGF-I and IGF-I), myog...

Wilson’s Disease: Nutrition Support

Wilson’s disease (WD) or hepato-lenticular degeneration is a rare autosomal-recessive disorder. A prevalence rate of 30 cases per million and a birth incidence rate of one per 30,000 to 40,000 are often quoted. In 40 to 50% of individuals with WD, hepatic dysfunction is the initial clinical manifestation. With the exception of liver transplantation, treatment of WD is only palliative and intend...

Effects of Environmental parameters on Metallotheionin changes in polycheate (Glycinde boundari) of Chabahar Bay

In addition to heavy metals, environmental factors affect to induce the synthesis of metallothionein (MT) contamination biomarker in vertebrates and invertebrates. Environmental parameters that were selected of nine stations in Chabahar Bay subtidal area (2013-14)  include temperature, salinity, dissolved oxygen and turbidity and heavy metals (copper, zinc, cadmium) in monsoon and post-monsoon,...

Interleukin-6 regulates the zinc transporter Zip14 in liver and contributes to the hypozincemia of the acute-phase response.

Infection and inflammation produce systemic responses that include hypozincemia and hypoferremia. The latter involves regulation of the iron transporter ferroportin 1 by hepcidin. The mechanism of reduced plasma zinc is not known. Transcripts of the two zinc transporter gene families (ZnT and Zip) were screened for regulation in mouse liver after turpentine-induced inflammation and LPS administ...
